【摘要】:Bacterialpopulations,aspartofthemicrobialcommunityinaquaticsystemsareactivelyinvolvedinnutrientcycling.Thesignificanceofthesepopulationsisoftendifficulttoascertainbecauseofthepresenceofmanyphysiologicaltypes.However,measurementofbacterialdensitiesisusuallythefirststepintryingtoestablishanyrelationshipthatmightexistbetweenbacteriaandotherbiochemicalprocesses.Acridine-orangeepifluorescencedirect-countingprocedurecannotdifferentiatebetweenviableandnonviablecells.Thisprocedurecannotbeusedtoconvertdirectlythenumberstototalcarbonbiomassbecauseofthenaturalvariationsinbacterialcellsize.Theacridine-orangeepifluorescencedirect-microscopiccountisbothquantitativeandprecise.Thisprocedureisidealforenumeratingbothpelagicandepibenthicbacteriainallfreshwaterandmarineenvironments.Theprocesscanbeemployedinsurveyactivitiestocharacterizethebacteriologicaldensitiesofenvironmentalwaters.Theprocedurecanalsobeusedtoestimatebacterialdensitiesincoolingtowerwaters,processwaters,andwatersassociatedwithoildrillingwells.1.1Thistestmethoddescribesaprocedurefordetectionandenumerationofaquaticbacteriabytheuseofanacridine-orangeepifluorescencedirect-microscopiccountingprocedure.Itisapplicabletoenvironmentalwaters.1.2Certaintypesofdebrisandothermicroorganismsmayfluoresceinacridineorange-stainedsmears.1.3Thetestmethodrequiresatrainedmicrobiologistortechnicianwhoiscapableofdistinguishingbacteriafromotherfluorescingbodiesonthebasisofmorphologywhenviewedathighermagnifications.1.4Useofbrightlightpermitsdifferentiationofsinglebacteriawherereducedformazanisdepositedatthepolarends.1.5Approximately104cells/mLarerequiredfordetectionbythistestmethod.1.6ThevaluesstatedinSIunitsaretoberegardedasstandard.Nootherunitsofmeasurementareincludedinthisstandard.1.7Thisstandarddoesnotpurporttoaddressthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
